Efeito do hormônio 17β-estradiol na gametogênese dos órgãos de Bidder e testículos e no tecido hepático em machos de Rhinella schneideri (Anura: Bufonidae)

Abstract

O declínio das populações de anfíbios é um problema global com causas complexas. Atualmente há uma grande preocupação sobre os efeitos provocados por compostos que atuam como desreguladores endócrinos nos sistemas naturais. Os estrogênios são os principais contaminantes encontrados em corpos d´água e são responsáveis por causar uma série de distúrbios, principalmente ligados aos aspectos reprodutivos. O hormônio feminino 17β-estradiol é um potente contaminante que prevalece em águas superficiais, provenientes principalmente de efluentes municipais e industriais. É o hormônio responsável pela formação das características femininas, comportamento sexual e ovulação. Este estudo mostra o efeito do 17β-estradiol na gametogênese dos órgãos de Bidder e testículos e na resposta hepática de machos de Rhinella schneideri. Foram utilizados 40 machos adultos e analisada a gametogênese dos testículos e órgão de Bidder, além da avaliação da pigmentação hepática. Os animais foram tratados com 17β-estradiol durante 3, 7, (efeito de curto prazo) 15 e 30 (efeito de exposição prolongada) dias, utilizando injeções subcutâneas de 2mg/kg em dias alternados. Análises histoquímicas também foram realizadas para os três órgãos. Os resultados mostraram que o 17β-estradiol promoveu alterações tanto na espermatogênese, quanto na ovogênese bidderiana. Os tratamentos de curto prazo acarretaram em uma diminuição na quantidade de espermatozoides liberados no lume e um desequilíbrio na ocorrência de células iniciais da espermatogênese. Os tratamentos de exposição prolongada acarretaram principalmente na diminuição dos lóculos seminíferos e aumento na região intersticial, além da alteração de alguns tipos celulares como espermatogônias, espermatócitos e espermátides alongadas. Amphibian’s populations decline is a global problem with complex causes. Currently, it is known that this decline is related to endocrine disruptors compounds in natural systems. Estrogens are the main compounds found in water bodies and are responsible for causing a number of disturbances, mainly linked to reproductive aspects. The female hormone 17β-estradiol is a potent organic contaminant that prevails in surface waters, mainly disposed by municipal and industrial effluents. This hormone induces the formation of feminine characteristics, sexual behavior and ovulation. In this study, we describe the effect of 17β-estradiol in the gametogenesis of the Bidder's organ and testes, beyond the liver response in males of Rhinella schneideri. We collected 40 adult males and analyzed the testis and the Bidder´s organ gametogenesis and the liver pigmentation. The animals were treated with 17β-estradiol for 3, 7, (short effect) 15 and 30 (extended effect) days, by subcutaneous injection of 2mg/kg on alternate days. Histochemical analyzes were also performed for the three organs. The results showed that 17β-estradiol promoted changes in spermatogenesis and in bidderian oogenesis. Short-treatments resulted in a decrease in the quantity of spermatozoa in the lume and an imbalance in the occurrence of initial cells of spermatogenesis. Long-treatments resulted in a decreased of the seminiferous locules and increased of the interstitial area, besides the alteration of some cell types, such as spermatogonias, spermatocytes and elongated spermatids. In the Bidder´s organ, the short-treatment resulted in increased of the young follicles and degeneration oocytes, establishing a possible renewal of female cells.
